Effect of Penicillic Acid on Na-K-ATPase Activity of Rabbit Cerebral Cortex

Abstract
|
|
|
The kinetics of inhibition by penicillic acid of Na-K-ATPase activity was investigated with rabbit cerebral cortex.
1. The inhibition of Na-K-ATPase activity showed sigmoidal curve with 6 (the inhibitor concentration to cause 50% inhibition) of 3.8X10-¢¥ M.
2. The fractional inhibition by a constant concentration of penicillic acid increased with the diminishing enzyme concentration. This result suggests penicillic acid inhibition of Na-K-ATPase activity was irreversible.
3. Kinetic studies of cationic-substrate activation of Na-K-ATPase showed competitive inhibition for Na¢¥, while showing noncompetitive inhibition for ATP and K+.
4. Cumulative inhibition studies with penicillic acid and ouabain indicated possible interaction between the two inhibitors of Na-K-ATPase.
These results show that penicillic acid inhibits Na-K-ATPase activity by interacting at Na¢¥ sites, and the mode of action of peniciilic acid may be different from ouabain.
